Technical skills and competences During my research activity, I gained considerable experience in the manipulation of primary human endothelial cells and pericytes as well as the preparation of co-cultures that represent an elegant in vitro model of barrier. Co-cultures give the possibility of cultivating cell lines in close contact, in order to reproduce and mimic the characteristic of biological barriers and better define cell-cell interaction in both physiological and pathological conditions. All data collected so far indicate that there is a real mutual influence of both cell types in this co-culture condition. My research is focused on in vitro studies about blood-retinal barrier (BRB) or blood-brain barrier (BBB) models challenged with pro-inflammatory stimuli such as high glucose or pathogens in order to study molecular mechanisms or evaluate the effect of drugs. In these models, I assess the barrier integrity through the measurement of trans-endothelial electrical resistance (TEER), the diffusion of sodium fluorescein molecules or tight junction protein expression. I currently carry out cell viability test (MTT assay), proliferation tests (Trypan Blue exclusion test or BrdU assay), tube formation assay and cell toxicity test (LDH release). I performs protein expression studies by western blot, ELISA evaluation of inflammatory cytokines and studies of mRNA expression by real-time PCR. I prepare also co-cultures of endothelium and pericytes in presence of tumor cells (melanomas or gliomas) as very elegant triple-culture model. I use light optical microscopy for cell migration assays (wound-healing assays) and time-lapse proliferation, as well as fluorescence or confocal microscopy to study proteins distribution and expression. Through my PhD, I have expanded my experience in the field of cloning using T4 DNA ligases, plasmid DNA preparations, transfection and luciferase assays.
