In vitro permeation of aceclofenac through the shed skin of two different species

Saponins rich fraction of Caralluma adscendens, Gymnema sylvestre & Piperine, an amide alkaloid of black pepper, was investigated for transdermal enhancer activity using shed snake skin in vitro with aceclofenac as the model drug. The membranes used were the dorsal portions of shed skin of two different species of snake. Results shows excellent permeability in dorsal area with permeability order for different species is cobra > viper, this may be due to thickness of skin, significantly increased flux of the drug compared to control (p<0.05). Similarly, permeability coefficient (Kp), cumulative amount release (Q24) and enhancement ratio (ER) shown significant increase over control sample. These results indicate that Caralluma, Gymnema & Piperine enhances transdermal permeation of aceclofenac significantly. The combination of two enhancer (C+G) showed most promising result.
