The goal of the proposed research is to investigate the regulation of Ikaros leukemia suppression activity. Specifically, we focus on understanding how Ikaros anti-tumor activity is modulated by Casein Kinase II inhibition and epigenetic regulation on Ikaros targets in primary leukemia, and will provide solid evidence for combination of CKII inhibitor with inhibitor of oncogenic pathways as a therapy on ALL leukemia. I have a broad background in doing research on regulation of gene?s function, signaling pathways, and transcriptional and epigenetic regulation on leukemogenesis and hematopoiesis. My current research is focused on leukemia. I worked on the regulation of Ikaros function by Casein Kinase II (CK2) and Protein phosphatase. I identified genome-wide chromatin state of Ikaros and HDAC1 targets in human leukemia cells, which suggests epigenetic regulation and plays a key role in modulation of function of Ikaros targets. One paper concerning these studies is published on Blood recently and another one is just accepted by ?Oncotarget?. Also the 3rd one is revising for ?Leukemia?; these studies not only built the basis for the proposed research but also established required methods and techniques for successful completion of the proposed aims in this proposal. Significantly, so far this year, I have published 6 papers. Two of them is 1st author paper on Blood and Leukemia, co-1st author paper on J Biol Chem, 3 of them as correspondence author on Oncotarget (2 papers) and Clin Microbiol Infect (1 paper); in addition, 1 manuscripts as correspondence author is submitted for Leukemia. My working experience on Ras and phospholipid signaling will also benefit the proposed research on function of Ikaros genes of multi-congenic pathways and drug resistance genes. As a PhD student at Kobe University in Japan, I identified a novel phospholipase C (named as PLC?) which is regulated by Ras oncogene. As a postdoctoral fellow at UW-Madison, I worked on regulation of phospholipids on RNA processing, and as key researcher I identified a novel Poly (A) polymerase (Star-PAP). As collaborator, I also worked on the anti-tumor effect of adenosine by inducing apoptosis and senescence, and also ion channel which is involved in drug metabolism in cancer cells. In addition, I graduated from medical school, got the residence training on clinical medicine in China, and have a clinical Doctor as my mentor which grantee the completion of the project concerning primary leukemia. Also, I have plenty of working experience for managing projects, collaborating with other researchers. In summary, I am well qualified to undertake the studies proposed, have the materials and skills to carry out the work, and the experience to deal with unforeseen opportunities or problems as they arise during this work.
Research Interest
The goal of my research is to understand the epigenetic and transcriptional regulation of cancer and develop novel therapy for cancer. Currently, my research focuses on childhood leukemia, particularly identification of genome-wide chromatin state of Ikaros and HDAC1 targets in primary human leukemia cells. We found that targeting casein kinase II restores Ikaros tumor suppressor activity and demonstrates therapeutic efficacy in High-Risk leukemia. By collaboration with other researchers, I also identified miR-215 is a bio-marker in human glioma, c-MYC, LEF1 and CRLF2 in human leukemia, and also analyzed the clinical significance of the bio-markers. My contributions to science include transcriptional and epigenetic regulation in Leukemia and CK2 inhibitor for leukemia therapy, identification of novel poly A polymerase regulated by phospholipid (Star-PAP), discovery of a novel phospholipase C (PLCepsilon) regulated by Ras, identification of biomarkers in cancer, anti-tumor effect of potential new drugs, and work on ion channels, metabolism and hepatitis infection.
