Abstract

A Cytochrome 450 Interaction of Chloroquine with Second Generation Antipsychotics

The temporal association of seizure as an adverse drug event is extremely rare with therapeutic doses of chloroquine as evident from its inception and widespread use in clinical treatment of malaria. However, chloroquine has potential to interfere in the metabolism of clozapine and risperidone via CYP 450 enzyme system; both being synergistically used in treatment of treatment resistant schizophrenia. This case illustrates the isolated case of possible chloroquine induced convulsive seizure in patients of schizophrenia, who was stabilised on clozapine 200 mg per day and risperidone 6 mg/day and discusses the interactional mechanisms therein.
